                | I. Background | |
| 
				       4-1BB [也稱為 CD137 或 TNF 受體超家族成員 9 (TNFRSF9)] ,是糖基化的 I 型膜蛋白,屬于 TNFR 超家族成員,由四個(gè)胞外富含半胱氨酸的假重復(fù)結(jié)構(gòu)域組成。4-1BB 的胞質(zhì)區(qū)包含 TNF 受體相關(guān)因子 (TRAF) 結(jié)合基序。4-1BB 表達(dá)主要發(fā)現(xiàn)于活化的細(xì)胞毒性CD8 + T 細(xì)胞和輔助性 CD4 + T 細(xì)胞的表面,活化后,NK 細(xì)胞、B 細(xì)胞、單核細(xì)胞和樹突狀細(xì)胞中可誘導(dǎo) 4-1BB 表達(dá)。作為可誘導(dǎo)的共刺激分子,4-1BB 可增強(qiáng) T 細(xì)胞對抗原的反應(yīng)。在 T 細(xì)胞上被 4-1BBL 上調(diào)和三聚化后,4-1BB 會(huì)募集 TRAF 銜接蛋白至細(xì)胞漿TRAF 結(jié)合基序,從而啟動(dòng)共刺激信號(hào)傳導(dǎo)。 
				      4-1BBL (TNFSF9) 是 4-1BB 的主要配體,主要在 APC 、B 細(xì)胞、巨噬細(xì)胞和其他細(xì)胞類型中表達(dá),并與 4-1BB 結(jié)合,通過 TRAF1 和 TRAF2 誘導(dǎo)信號(hào)傳導(dǎo),從而激活 NF-κB、AKT、p38 MAPK 和 ERK 通路。NF-κB 誘導(dǎo) BCL-XL、BCL-2 和 c-FLIP 的轉(zhuǎn)錄,并刺激IL-2、IL-4 和 IFN-γ 的表達(dá),從而促進(jìn) T 細(xì)胞的存活和細(xì)胞毒性活性。因此,4-1BB/4- 1BBL 信號(hào)通路可以促進(jìn)不同類型的細(xì)胞存活。
