| I. Background | |
| GPR52 是一種 A 類(lèi)孤兒 G 蛋白偶聯(lián)受體（GPCR），尚未明確其內(nèi)源性配體，在中樞 神經(jīng)系統(tǒng)（尤其是紋狀體、伏隔核及前額葉皮層）高表達(dá)。其信號(hào)通路主要通過(guò)偶聯(lián) Gs 蛋白激活 cAMP/PKA 通路，促進(jìn) CREB 介導(dǎo)的基因轉(zhuǎn)錄，同時(shí)涉及β-arrestin 信號(hào)分支。作 用機(jī)制上，GPR52 與多巴胺 D2 受體共表達(dá)于紋狀體神經(jīng)元，通過(guò)調(diào)控多巴胺釋放及受體 活性，改善精神分裂癥陽(yáng)性癥狀（如幻覺(jué)）和運(yùn)動(dòng)障礙；在亨廷頓病中，GPR52 的激活可 抑制突變亨廷頓蛋白（mHTT）毒性，具有神經(jīng)保護(hù)潛力。 | |
